Click on the New Spot icon on the toolbar to start adding your own spot colours and then saving it under a spot table name.
 
 
As can be seen there are two modes for defining the spot colours, Lab and CMYK. The Lab colours can either be entered manually or using a colour measurement device such as XRite EyeOne. The CMYK method can only be done manually.
